Description

A method and kit to detect porcine in food/non-food items by utilizing isothermal amplification technique. The method includes the step of extracting DNA from the item. Then the method includes the step of quantifying the extracted DNA by utilizing picoGreen dsDNA quantitation kit. Further the method includes the step of utilizing pre-defined amount of DNA in an assay containing about 1-5 ?M of plurality of primers in presence of DNA polymerase and dye. The plurality of primers include mixture of primers with a nucleotide sequence in a direction from 5? to 3? end. Further the nucleotide sequence is derived from specific nucleotide region of porcines mitochondrial NADH dehydrogenase subunit V gene. Further, the method includes the step of carrying out a reaction at 60 degree Celsius for about 20 minutes - 40 minutes. Furthermore, the method includes the step of monitoring amplification of the reaction by utilizing fluorescence device.
