Description

Depression is the leading neuropsychiatric disorder that significantly contributes to the global burden of disease. Conventional antidepressants have not been sufficiently effective and they also produce many life-degrading side effects. So, the synergism effect of Lignosus rhinocerotis, Hericium erinaceus and Ganoderma Lucidum which are formulated into NevGro® forte have been reported to produce therapeutic potential for treating depression. The present studies exhibited that four-weeks of NevGro® Forte treatment ameliorated depressive-like behaviours in chronic unpredictable mild stress (CUMS) rat model. Significant weight improvement, brain's weight, and increased thickness of pyramidal layer in the hippocampus following the treatment of NevGro® Forte. Scanning electron microscopy also revealed decreased degeneration characterised by flattened with less dense surface composition in the hippocampus. Our histological brain analysis showed a thickness reduction on granular cell layer (GCL) in dentate gyrus (DG), in the negative group (CUMS-induced) as well as smaller pleomorphic layers (PL). Clinically, depressed patients had a 5% reduction in DG volume bilaterally, as well as lower CA and subiculum volumes when compared to healthy controls.

Highlights

1. Combination of three medicinal mushrooms (NevGro® Forte) may be a promising alternative medication for the treatment of depression based on the neurotrophic and neurogenic pathophysiology of depression. 

2.Furthermore, we also observed that pyramidal cell layer (PCL) of negative group (Chronic Unpredictable Mild Stress-induced) is in scattered arrangement as well reduced in thickness compared to the normal one. 

3. The results exhibited that 4 weeks of NevGro® forte treatment ameliorated depressive-like behaviours subjected to 56 days of chronic mild stress (CMS). Further molecular protein assays demonstrated the 4-week treatment elevated the serotonin level and reduced corticosterone level while elevating neurogenesis-related proteins, including brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF) and phosphorylated cAMP response element-binding protein (CREB). Moreover, histological studies also revealed decreased degeneration characterised by eosinophilic pyknotic neurons and Increased thickness of pyramidal layer in the hippocampus. NevGro® forte exerts antidepressant effects by promoting neurogenesis by enhancing the BDNF-CREB signalling pathway. 

4. In behavioural analysis: Significant differences were observed between the negative control (CUMS) and treatment groups upon completion of 28 days treatment with NevGro® Forte. It showed significantly reduce depressive-like behaviours and anxiety after oral administration of NevGro® Forte. 

5. A reduction in hippocampus volume is one neurological difference strongly related with serious depression in adults, we observed that the gross morphology, of the negative control (CUMS-induced) brain was slightly smaller compared to other groups where the mean volume and weight, is significantly low too. Besides that, the treatment groups of rats show significant values with the normal group of experimental rats, especially the treatment and pre-treatment groups which both uses NevGro® Forte are nearly corresponding to the normal group. 

6. Externally, it is characterized by a layer of densely compacted neurons that folds into an S-shaped structure which generates some of the greatest EEG waves that are related to learning and memory. The administration of NevGro® Forte at different concentrations have significantly improve the density surface with a compact topography. 

7. The production of this product does not involve harmful chemicals and does not produce manufacturing waste that is harmful to the environment. The 100% natural ingredients of this product ensures 0 negative impact on the environment.
