Disclosed is a reverse genetics system for producing a genotype-matched live attenuated vaccine against a genotype VII Newcastle disease virus (NDV). The reverse genetic system comprises a full-length antigenomic clone, helper plasmids, mammalian cells, and SPF eggs. The full-length antigenomic clone comprises an antigenome of NDV IBS025/13. The antigenome of NDV IBS025/13 is modified, synthesized and cloned into a low copy number pOLTV5-phix transcription vector. The helper plasmids vector the antigenome of NDV IBS025/13. The helper plasmids are expression constructs carrying an NP gene, a P gene and an L gene derived from the antigenome of NDV IBS025/13. Further, the helper plasmids are expressed in mammalian cells. The mammalian cells are BHK-21 cells genetically engineered to express a T7 RNA polymerase. The antigenome of NDV IBS025/13 is rescued following the expression of the T7 RNA polymerase with the full-length antigenomic clone and the helper plasmids. The SPF eggs amplify the rescued antigenome of NDV IBS025/13 from the mammalian cells.
