The present invention relates to a method for semen cryopreservation of tor spp., namely tor tambroides and tor douronensis and in vitro fertilization using the cryopreserved semen. The cryopreservation method comprises the steps of: obtaining semen from selected fish; diluting the semen using a diluent comprising an extender and a cryoprotectant, wherein the extender comprises d(+)-glucose monohydrate, sodium chloride and sodium bicarbonate while the cryoprotectant is dimethyl sulfoxide or methanol; packaging the semen; subjecting the semen to equilibration; and freezing the semen. The in vitro fertilization method comprises the steps of: obtaining unfertilized eggs; gently mixing the thawed cryopreserved semen with the unfertilized eggs; and adding a fertilization buffer into the semen and eggs mixture, wherein the fertilization buffer comprises sodium chloride, sodium bicarbonate and fructose.
