Allopurinol, carbamazepine, lamotrigine, phenobarbital, phenytoine, sulfamethoxazole, oxicam and nevirapineare are five high-risk drugs that could induced cutaneous adverse drug reaction, such as Stevens-Johnson syndrome and toxic apidermalnecrolysis. The current method of detection for these alleles utilizes expensive commercial genotyping kits to genotype the HLA groups or time consuming conventional gel-based PCR. There is a need to develop a rapid and cost effective screening method to detect these specific alleles, so that these pharmacogenetic screening can be made available in our community hospital setting. This project aims to develop a detection kit to screen for the alleles using the high resolution melt (HRM) technique which is time saving and cost effective compared to conventional PCR, conventional Real-Time PCR and commercial HLA genotyping kit. This method will also utilize a validated positive control which reduces the need to validate the result by DNA sequencing.
